Pressure-based sample preparation, including rapid enzymatic proteolysis and enhanced protein extraction and recovery, is now playing a key role in the field of proteomics.  Pressure Cycling Technology (PCT) is capable of reducing digestion times from hours to minutes, increasing the recovery of important proteins, and adding an additional level of standardization to the important sample preparation workflow of mass spectrometry and other downstream analytical methods.

The PCT-HD System combines two of PBI’s unique products, PCT μPestles and an enhanced Barocycler NEP2320.  PCT-HD was developed by PBI scientists and engineers,in collaboration with Dr. Ruedi Aebersold, his colleague Dr. Tiannan Guo.  Drs. Aebersold and Guo include the PCT-SWATH System as an integral part of PCT-SWATH in their research, publications, and presentations. Their term (PCT-SWATH) derives from the fact that they coupled the PCT-SWATH System with mass spectrometry and SWATH Acquisition (Sciex) to reproducibly quantify thousands of proteins from minute tissue biopsy samples (e.g. small needle biopsies).  This protocol can yield analytical results within 12 hours from the start of processing tissue.  However, the PCT-SWATH System and protocols developed by the Aebersold Lab are not limited to a specific mass spectrometer or other methods of data analysis.The PCT-HD can provide most researchers with unprecedented speed and reproducibility for biomarker discovery.

The Barozyme HT48 is a bench-top, high throughput PCT-based instrument designed specifically for accelerated enzymatic digestion of proteins at high pressure and elevated temperature. The Barozyme HT48 is capable of processing up to 48 samples simultaneously in six single-use BaroFlex 8-well Strips. The Barozyme HT48 uses the universally accepted "microplate" format, allowing the system to integrate with robotic automation found in today's modern research laboratories. With the Barozyme HT48, typical trypsin digestion times can be reduced from hours to minutes, while enjoying the advantages of increased quality in sample preparation that is intrinsic with PCT.
